HPLC System Suitability Calculator

Calculate chromatographic resolution, theoretical plate number, peak tailing factor and percent relative standard deviation.

HPLC system suitability calculations

These forms calculate values only. They do not set acceptance criteria or determine whether a method is suitable.

Resolution between two peaks

Select the convention used by your procedure. Use matching time units for retention times and widths.

Theoretical plate number

Use the same time unit for both values.

Peak tailing factor

Use the same width unit for both measurements.

Percent relative standard deviation

Enter at least two positive finite results, separated by commas or new lines.

In short: Calculate common HPLC system suitability values from measured retention times, peak widths or replicate results. This tool calculates values only; acceptance criteria come from the applicable monograph or validated procedure.

Calculations

Resolution

The calculator offers two peak-width conventions. For baseline widths, it calculates:

Rs = 2 × (tR2 − tR1) ÷ (W1 + W2)

For widths measured at half-height, it calculates:

Rs = 1.18 × (tR2 − tR1) ÷ (Wh1 + Wh2)

Use the convention specified by the applicable method and make sure both retention times and widths use the same time unit. The harmonized chromatographic convention uses widths at half-height; the baseline-width equation is retained here for comparison with legacy procedures and the stated reference calculation. USP General Chapter <621>, Chromatography (2024); Ph. Eur. General Chapter 2.2.46, Chromatographic separation techniques, 11th Edition (2023).

Theoretical plate number

N = 5.54 × (tR ÷ Wh)²

Here, tR is retention time and Wh is peak width at half-height. Use the same time unit for both inputs. The displayed calculation is a column-efficiency measure; method requirements determine whether a particular result is acceptable. USP General Chapter <621>, Chromatography (2024).

Peak tailing factor

T = W0.05 ÷ (2f)

W0.05 is peak width at 5% of peak height. f is the distance from the peak apex to the leading edge at that same height. Both measurements must use the same unit. USP General Chapter <621>, Chromatography (2024).

Percent relative standard deviation

s = √[Σ(xi − x̄)² ÷ (n − 1)]
%RSD = 100 × s ÷ x̄

This calculator uses the sample standard deviation (n − 1) and requires at least two positive replicate results. The tool does not apply a limit to the calculated %RSD. USP General Chapter <1010>, Analytical Data—Interpretation and Treatment (2021).

Assumptions and limitations

  • No acceptance limit, injection count or pass/fail status is built in. Follow the individual monograph or validated procedure.
  • Enter compatible units within each formula. For resolution, select the width convention matching the method.
  • The calculator does not read a chromatogram, choose peaks, correct integration or judge whether the system is suitable.
  • Results are arithmetic aids and do not replace a validated analytical procedure or review of the chromatographic data.
