In short: PIC/S published PI 006-4, revised Recommendations on Qualification and Validation, on 30 July 2026. It replaces PI 006-3 (2007) and enters into force on 1 October 2026.

What changed

PI 006-4 is a 49-page document that, in its own words, updates and renames PI 006-3. The older document was titled for four topics: validation master plan, installation and operational qualification, non-sterile process validation and cleaning validation. The revision history in PI 006-4 gives the reason for the update: alignment with the revised chapters 3 and 5 and with Annex 15 of the GMP Guide.

The document now has a broader structure:

  • Introduction (purpose, scope, aims, terminology, change control, organisation and responsibilities, quality risk management)
  • Validation master plan
  • Prequalification and qualification stages (IQ, OQ, PQ, re-qualification) for facilities, utilities, equipment and systems
  • Process validation: prospective, concurrent, approaches to support initial commercialisation, ongoing (continued) process verification, revalidation
  • Other topics: verification of transportation, validation of packaging (solid dose only), qualification of supporting utilities, validation of test methods
  • Cleaning validation (principle, documentation, personnel, equipment, microbiological aspects, visual inspection, sampling, detergents, analytical methods, limits, maintaining a validated process)
  • Glossary and revision history

Section 2.5 states that qualification and validation activities should be conducted using a lifecycle approach, appropriate to the lifecycle stage of the facility, utility, equipment, system or process. Section 2.8 ties the scope and extent of the work to a justified, documented risk assessment.

The document says it applies to APIs, intermediates and finished dosage forms alike. It does not cover computerised system validation (Annex 11), does not treat analytical method validation in depth, and leaves topics covered elsewhere, such as aseptic process validation (PI 007).

PIC/S notes that the technical text was prepared by a dedicated working group, first chaired by Norman Gray (UK/MHRA) and later co-chaired by Kevin O’Donnell (Ireland/HPRA) and Rebecca Dowd (USA/FDA). The document itself does not give a list of line-by-line differences from PI 006-3.

Who it affects

PI 006 is guidance for GMP inspectors and industry, not a law. It states that it serves as a training resource for inspectors and as a reference for a GMP-compliant validation programme, so firms inspected by or exporting to PIC/S authorities should know it.

Key dates

  • 25 September 2007: PI 006-3 entered into force.
  • 30 July 2026: PIC/S announcement.
  • 1 October 2026: PI 006-4 enters into force.

What to do now

  • Read PI 006-4 against your validation master plan, qualification, process validation and cleaning validation procedures.
  • Update document references from PI 006-3 to PI 006-4.
  • Check whether your documents take a lifecycle and risk-based approach (example in-house approach: a short gap table by section, owners and due dates).
